Nikki Tamboli belongs to a Christian family from Aurangabad, Maharashtra. Her father, Anil Tamboli is a sales executive in a bank and her mother, Mukti Tamboli is a homemaker.
She completed her schooling from Aurangabad and her higher studies from Kishinchand Chellaram College. Tamboli started her career as a model and has appeared in several commercials on television.

Tamboli has appeared in films like Kanchana 3, Thippara Meesam, and Chikati Gadhilo Chithakkotudu. She made her acting debut in 2019 from a Telugu horror-comedy, Chikati Gadilo Chithakotudu, which was directed by Santhosh P. Jayakumar. It was a remake of the Tamil hit Iruttu Araiyil Murattu Kuththu. She then went on to make her Tamil debut with the blockbuster Kanchana 3 alongside Lawrence. She also appeared in the Telugu thriller, Thippara Meesam.
